For XP or Vista
Open My Computer.
Right-click the local disk (drive) that you want to defragment, and then click Properties.
On the Tools tab, click Defragment Now.
Click Defragment.

You are better of not working on the computer whilst it is doing the defrag. Depending of the size of the drive, be prepared for a wait of up to a couple of hours while it sorts it out if you have not done it for a long time.
